应用多重PCR技术检测性器官异常患者的性别决定基因

采用针对人SRY基因及X与Y染色体同源序列的两对引物进行多重聚合酶链反应技术检测204例新生儿脐血DNA,男性均显示590bp、 355bp及280bp 3条扩增带,女性只有590bp1条扩增带,性别鉴定准确率为100%。又检测47例性器官异常患,13例社会性别为女性者只出现590bp扩增带,与核型性别结果一致。 34例社会性别为男性者中,有2例SRY基因检测结果阴性,只出现590bp带,核型为46,XX,证明这两例患者社会性别不相符,经病理证实后应诊断为女性假两性畸形。 Abstract:This investigation adapted multiplex PCR technique with two pairs of primer to determine sex according to the human SRY gene and X,Y chromosome analogical sequence.We detected bellybutton blood DNA from 204 newborns.Male revealed three bands:590bp,355bp and 280bp,and females only had one band which was 590bp,the accuracy of sex determination was 100%.47 sexual abnormal patients were tested.The results showed that 13 cases,whose social sex were female,had one 590bp band which consistent with their chromosome type sex.In the rest 34 cases,whose social sex were male,2cases showed,on the contrary,only one 590bp band and their chromosome type were 46,XX.This proved that the two patients did not consistent with their social sex,and pathological analysis showed that they were female pseud-hermaphroditism.

普通小麦品种Hope细胞膜热稳定性基因的染色体定位

利用全部 21个“中国春”的“Hope”染色体代换系及其亲本品种“中国春” (受体)和“Hope”(供体)对六倍体普通小麦的细胞膜热稳定性基因进行了染色体定位研究。结果表明:普通小麦品种“Hope”的1A、2A、2B、 2D、3A、3B、3D、5D和6B等9条染色体上具有耐热性基因,而其余染色体与“Hope”的耐热性无关。 Abstract:All 21 substitutions of common wheat(T.aestivum L.)and their parental cultivars“Chinese Spring”(recipient)and “Hope”(donor)were evaluated for their relative heat tolerance as measured by membrane thermostability to determine the chromosomal locations of genes controlling this trait.Results indicate that chromosomes 1A,2A,2B,2D,3A,3B,3D,5D and 6B were associated with heat tolerance of cv.Hope,while the others were not related to heat tolerance.     

The first mitochondrial genome for the butterfly family Riodinidae(Abisara fylloides) and its systematic implications

The Riodinidae is one of the lepidopteran butterfly families. This study describes the complete mitochondrial genome of the butterfly species Abisara fylloides, the first mitochondrial genome of the Riodinidae family. The results show that the entire mitochondrial genome of A. fylloides is 15 301 bp in length, and contains 13 protein-coding genes, 2 ribosomal RNA genes, 22transfer RNA genes and a 423 bp A+T-rich region. The gene content, orientation and order are identical to the majority of other lepidopteran insects. Phylogenetic reconstruction was conducted using the concatenated 13 protein-coding gene(PCG) sequences of19 available butterfly species covering all the five butterfly families(Papilionidae, Nymphalidae, Peridae, Lycaenidae and Riodinidae). Both maximum likelihood and Bayesian inference analyses highly supported the monophyly of Lycaenidae+Riodinidae,which was standing as the sister of Nymphalidae. In addition, we propose that the riodinids be categorized into the family Lycaenidae as a subfamilial taxon.     

禁食与胰岛素处理对鳜血糖和leptin-A基因表达的影响

为研究硬骨鱼类leptin基因表达与血糖之间的关系, 研究在禁食与胰岛素处理后, 检测鳜血糖和肝脏、肠道和脂肪组织leptin-A基因表达水平。在禁食实验中, 鳜被禁食10d, 并分别于禁食后0、4h、2d、6d和10d取样。禁食后6d鳜血糖开始降低, 禁食后10d鳜血糖显著降低。同时, 禁食后6d鳜肝脏leptin-A基因mRNA表达水平显著升高, 禁食后4h鳜肠道和脂肪组织leptin-A基因mRNA表达水平均显著升高。在胰岛素处理实验中, 分别于注射后12h和36h取样。腹腔注射胰岛素后12h, 鳜血糖显著降低, 而鳜肝脏、肠道和脂肪组织leptin-A基因mRNA表达水平无明显变化。腹腔注射insulin后36h, 鳜肠道leptin-A基因mRNA表达水平显著升高。研究结果表明, 长期禁食或胰岛素处理均能够降低鳜血糖水平, 且影响消化器官和脂肪储存器官leptin-A基因表达。     

Immune Responses and Protective Efficacy Induced by 85B Antigen and Early Secreted Antigenic Target-6 kDa Antigen Fusion Protein Secreted by Recombinant Bacille Calmette-Guérin

In an attempt to improve immune responses and protective efficacy, we constructed two recombinant bacille Calmette-Guérin （rBCG） strains expressing an 85B antigen （Ag85B） and early secreted antigenic target-6 kDa antigen （ESAT6） of Mycobacterium tuberculosis （MTB） fusion protein. Both rBCG strains have the same protein insertion but in a different order （Ag85B-ESAT6 and ESAT6-Ag85B）. The cultured supernatant of rBCG strains and the sera from the mice immunized with the fusion protein Ag85B- ESAT6 or ESAT6-Ag85B formed a band with a fraction size of 37 kDa, equalivalent to the sum of Ag85B and ESAT6. Six weeks after BALB/c mice were immunized with BCG or rBCG, spleen lymphocytes showed significant proliferation in response to culture filtrate protein of MTB. Compared with the BCG group, mice vaccinated with rBCG elicited a high level increase of immunoglobulin G antibodies to culture filtrate protein in the serum. The T-interferon levels in the lymphocyte culture medium supernatants increased remarkably in the rBCG1 group, significantly higher than that of the BCG immunized group （P〈0.05）. Four weeks after vaccination, mice were infected with M. tuberculosis H37Rv and a dramatic reduction in the numbers of MTB colony forming units in the spleens and lungs was observed in the two rBCG immunization groups. Although these rBCG strains were more immunogenic, their protective effect was comparable to the classical BCG strain, and there were no significant differences between two rBCG groups （p〉0.05）.     

Calcium dobesilate for diabetic retinopathy: a systematic review and meta-analysis

Many randomized clinical controlled trials have confirmed the efficacy and safety of calcium dobesilate in treating diabetic retinopathy(DR).This systematic review critically evaluated the evidence that links calcium dobesilate to DR.In this fixed-effects meta-analysis,a total of 221 pertinent English-language articles published between January 1975 and October 2013 were identified.Systematic searches of PUBMED,Springer Link and the Cochrane Clinical Trials Database were conducted using the keywords “diabetic retinopathy” and “calcium dobesilate”.The extracted information included the study design,inclusion and exclusion criteria,setting,sample size,participant mean age,treatment regime,mean change in best corrected visual acuity,laboratory parameters,capillary fragility,intraocular pressure and fundus manifestations based on the findings of fluorescent angiography.The summary statistics indicated that calcium dobesilate was significantly associated with improving retinal microaneurysms(RR: 0.62,95%CI: 0.42?0.90,P=0.01),retinalhemorrhages(RR: 0.39,95% CI: 0.17?0.88,P=0.02); exudates(RR: 0.31,95% CI: 0.12?0.81,P=0.02),reduction of whole blood viscosity(MD: ?0.57 CP,95% CI: ?0.75 to ?0.38,P<0.001),plasma viscosity(MD: ?0.36 CP,95% CI: ?0.63 to ?0.09,P=0.01) and blood cholesterol(MD: ?0.48 mg m L?1,95% CI: ?0.64?0.33,P<0.00001).Intraocular pressure was also significantly reduced(MD: ?5.59 mm Hg,95% CI: ?6.69 to ?4.50,P<0.00001).The results indicate that calcium dobesilate effectively treats DR at the systematic and local ocular levels.     

Virus genomes and virus-host interactions in aquaculture animals

Over the last 30 years,aquaculture has become the fastest growing form of agriculture production in the world,but its development has been hampered by a diverse range of pathogenic viruses.During the last decade,a large number of viruses from aquatic animals have been identified,and more than 100 viral genomes have been sequenced and genetically characterized.These advances are leading to better understanding about antiviral mechanisms and the types of interaction occurring between aquatic viruses and their hosts.Here,based on our research experience of more than 20 years,we review the wealth of genetic and genomic information from studies on a diverse range of aquatic viruses,including iridoviruses,herpesviruses,reoviruses,and rhabdoviruses,and outline some major advances in our understanding of virus–host interactions in animals used in aquaculture.
